제목 | Preparation and characterization of double-layered mesospheres using w/o/w emulsion |
초록 | For molecules with narrow therapeutic ranges or high toxicity, several of the drugs could be required controlled release. To eliminate initial burst of drug, mesospheres have been studied. mesospheres made with a two-layered structure may have certain advantages comparing with single layered spheres.1,2 In this study, the dextran(MW=43,000) and poly(D,L-lactide-co-glycolide) 50:50 (PLGA) composite mesospheres were fabricated by a water-in-oil-in-water (w/o/w) double emulsion process.3 Predetermined rates were 1:1, 1:2, 1:3, 2:1 and 3:1 (dextran:PLGA, w/w %). The morphology of mesospheres varied depending on rate of dextran and PLGA. Dextran was dissolved in 0.5㎖of PBS(pH 7.4, containing polyvinylalchol 2 w/v %). After dissolving PLGA in MC, two solutions mixed and sonicated for 5 sec. this solution added in PBS and then freeze dried. As a result, double-layered mesospheres (average size=10㎛) could be fabricated.(그림. 1) The result of HPLC has shown slow release rate. Fig. 1.
