A novel acetobacterium sp. ES2 strain, isolated from a reed wetland of Eulsukdo in Korea, is an strict anaerobic, acetogenic bacteria via autotrophic or heterotrophic metabolism. Because of this property, ES2 strain is considered one of promising bacteria for carbon neutral. In this study, culture medium of ES2 strain was optimized to maximize cell growth and acetic acid production based on response surface methodology (RSM) by using three factors, which are initial concentration of methanol, NaOH and yeast extract. As a result of RSM, cell and acetic acid concentration could be maximized to OD600 of 2.86 and 5.0g/L, respectively, when the initial concentrations of methanol, NaOH and yeast extract were 134mM, 26mM, and 9g/L, respectively. The prediction was validated by batch culture of ES2 under optimized conditions, and the resulting cell and acetic acid concentrations were measured to be OD600 of 2.42 and 4.42 g/L, respectively, with the consistency of 85~88%. |
