Zwitterionic polymers with high biocompatibility and extremely low folding properties have been attracted as a new material in preparation of nanocarriers. In this work, poly(sulfobetaine) (PSB) was synthesized via Atom transfer radical polymerization (ATRP) and conjugated with functionalized thiolactone group. Subsequently, by one-pot thiol-ene polymerization, the thiolactone ring was opened by aminolysis of cystamine and released thiol-group for reaction with double-bond to form nanogel network. Under the presence of GSH as reduction agent, the degradation of nanogels was investigated by particles size change (DLS) and surface morphology (TEM). Doxorubicin (DOX), a model anticancer drug, was loaded into to the core of the nanogels and obtained a high drug loading content of 24.3%.
