Optimization and Separation of Racemic Clenbuterol by Simulated Moving Bed |
초록 |
SMB(simulated moving bed) is a powerful technique in continuous chromatography. The operation costs including eluent and chiral stationary phase are usually lower than batch chromatography and the main advantage of the continuous system are higher yield and separation efficiency. Recently, It has been extended to the application of enantioseparation. Enantiomers share the same physical properties except optical property and have differently biological activity. R-Clenbuterol is used for the treatment of pulmonary diseases. Adsorption isotherm and kinetic parameters for SMB simulation from a single-column experiment was presented. Configuration of SMB unit was 2-2-2-2, and Kromasil was used chiral stationary phase. The mobile phase composition is hexane/2-propanl/ acetic acid/triethyl amine, 95/5/0.2/0.1 (v/v%). The continuous experiments were performed at the optimized operating conditions by a software of Aspen Chromatography with the estimated parameters. |
