제목 Efficient biosynthesis of chemicals from methane via reconstruction of multi-carbon utilizing pathway in obligate type I methanotrophic bacteria
초록 Methanotrophs have attracted much attention as a promising biocatalyst for direct conversion of methane into fuels and valuable chemicals. However, production of reduced chemicals and fuels is limited by not only the low efficiency of methane assimilation but also the unbalanced reducing equivalents used in methane oxidation by MMO. In order to address this issue, we engineered M. alcaliphilum 20Z for co-utilization of multi-carbon glycerol and methane to enhance bacterial growth and provide extra reducing power to production of reduced fuels and chemicals. To demonstrate this concept, a strain habouring 2,3-BDO biosynthetic pathway and a MDH-deficient strain were constructed, and it was confirmed that the titer of each product was improved compared to the methanotrophic mode by co-utilizing methane and glycerol. The work presented here expands on the substrate spectrum in methanotrophs, which could provide a new framework for industrial biomanufacturing via methane bioconversion.
