Fabrication and Characterization of PLGA Microspheres Incorporated Hydrogel Loaded Porous Scaffold for Controlled Drug Delivery and Bone Regeneration |

In this study porous BCP scaffold of was prepared by sponge replica process. Then PLGA microspheres loaded with Simvastatin, a drug that enhances bone regeneration, were prepared by solvent emulsification method. These microspheres were combined with Gelatin hydrogel and poured onto the scaffold so that both entered inside the scaffold. To ensure the stability of the hydrogel in aqueous environment it was crosslinked with EDC (3-(3-dimethyl aminopropyl) carbodiimide). Mechanical properties testing and in-vitro experiments were performed to characterize the novel scaffold and to evaluate the biocompatibility and the drug efficacy and measuring the drug release profile. In compressive strength testing mechanical strength of the BCP scaffold was found higher after adding hydrogel in dry condition. SEM and FT-IR data confirmed the presence of both PLGA microspheres and Simvastatin inside the scaffold. Live and dead assay by confocal imaging and MTT, showed no toxicity against the pre osteoblast cell MC3T3-E1 activity. Western Blot and Real-Time PCR data suggested favorable protein and gene expression by the scaffold system, respectively. The burst release of the drug from the microspheres was reduced significantly in addition to an almost steady state release for continuous four weeks. |
