Investigation on the phase development of Calcium phosphate powder depending on Ca/P ratio and heat treatment and comparison of the inherent biocompatibility of them |

Microwave hydrothermal synthesis process of Calcium phosphate powders like Hydroxyapatite and biphasic calcium phosphate is a high throughput synthesis method which produces mostly amorphous powders. The phase development from the amorphous synthesized materials is largely dependent on the post synthesis heat treatment of the powders and initial ratio of Calcium and Phosphate precursor. In this work we synthesized Calcium phosphate powders with Ca/P ratio of 1.2, 1.4. 1.6 and 1.8 and after synthesis exposed them at different calcination temperatures like 400, 600, 800 and 1000 ºC. Phase identification and characterization was carried out by XRD and FTIR. The phase development of the powders was analyzed depending on the corresponding Ca/P value and calcination temperature. Powder characteristics were further evaluated by SEM. To investigate the biocompatibility of the individual phases we performed an initial screening by MTT assay. The powders were suspended in Phosphate Buffer Saline and resulting pH was measured to evaluate the suitability of the different phases in physiological environment. Further biocompatibilities of the individual screened out phases were evaluated by cell proliferation behavior observed by SEM and confocal imaging and time dependent gene expression behavior by PCR. The results were used to index different phases of the microwave synthesized powders according to their comparative biocompatibility. |
