제목 Electro spun membrane wrapped BCP scaffold for selective ingrowth of osteogenic cells for total bone regeneration
초록 For larger than critical size bone defect, the bone healing is compromised by the invasive intervention of tissues other than the bone tissue. The bone scaffold mediates in the way of predominantly bone cell proliferation to complete the bone healing. Unwanted tissue growth needs to be protected for full bone regeneration. For regenerating large bone segment using porous scaffold this issues needs to be considered.  In our work we fabricated a cylindrical porous HAp scaffold with a central hole by sponge replica method, as a scaffold for large segmental bone replacement. The scaffold was wrapped with electro spun fiber mesh of biodegradable PLGA-Gelatin polymer composites to render the scaffold impregnable to lateral invasion of less desirable tissue ingrowth.  To facilitate the osteo-conduction and osteo-induction the scaffold was loaded with PLGA-Gelatin hydrogel. Detail morphological and material characteristics were evaluated by SEM observation and mechanical testing.  To investigate the biocompatibility of the fabricated system, selective cell proliferation behavior was observed by SEM and confocal imaging. Finally invivo model was used to evaluate the bone regeneration ability and bone tissue ingrowth selectivity. Tissue staining and immunohistochemistry analysis was carried out to elucidate the performance of the fabricated system.
