De novo biosynthesis of plants-specific stilbene compounds in E. coli |

Microbial synthesis of plant secondary metabolites has attracted increasing attention due to their proven beneficial properties and health-promoting effects. However, the difference between the metabolic pathways of microbes and plants is a challenging subject to build a de novo production system of metabolites in microbes. Stilbene, a type of natural phenol, is the precursor to a range of important plant metabolites such as resveratrol. Our strategy demonstrates the microbes for de novo synthesis of resveratrol analogs used a single vector system joined with resveratrol biosynthetic genes and other modified genes. In addition, most artificial de novo biosynthetic pathways are multi-reaction pathways that require a balanced expression of several biosynthetic enzymes. I will introduce our efforts in fine-tuning the expression level of a six-enzyme reaction for the artificial curcumin biosynthesis as a model system. |
