In this study, anticancer drug-conjugated multifunctional chitosan (AMC) was synthesized by hydrazone bond for pH-sensitive drug release system. In addition, Transferrin-AMC(TAMC) was prepared by coupling agent for specific tageting. Hydrazone bond can be disrupted to corresponding carbonyl bonding. Therefore, drug was released by hydrolysis of hydrazone bond in acidic condition. The characterization of TAMC was investigated by 1H-NMR and DLS. The drug release profile of AMC was confirmed by HPLC. Evaluation of in vitro cell cytotoxicity was confirmed by MTT assay. Confocal laser scanning microscopy (CLSM) was used to confirm the targeting efficiency and localization of TAMC. The TAMC proposed that the conjugated drug was easily released by hydrazone bond from endo/lysosome and effective to target tumor cells.
