제목 Development of gut-liver-kidney multi-organ chip to mimic ADEM of drugs
초록 New drug development can save time and money only when it is evaluated with efficiently selected new drug candidates in the preclinical stage. Therefore, preclinical models are important in drug development. However, animal experiments or human clinical trials, which are conventional preclinical models, are limited in terms of ethical issues and economics. As an alternative to the conventional preclinical trial model, an organ-on-a-chip was developed. Orally administered drugs pass through the gut-liver-kidney and undergo absorption-distribution-metabolism-excretion (ADME). To implement this process, a multi-organ chip in which the gut-liver-kidney is organically connected is required. On the other hand, the kidney is responsible for two roles, filtration (glomeruli) and reabsorption (tubules) functions, at the same time. To mimic these functions, it is necessary to additionally co-culture the cells responsible for each function. instead of glomerular cell, we added a glomerular film to the chip and observed the glomerular functionality. We hope that the chip could potentially be used as a new and more accurate preclinical model
