제목 DNA Aptamer-immobilized Hydroxyapatite for Enhancing Bone Regeneration and Angiogenesis
초록 Aptamers are short oligonucleotide structures that are capable of targeting molecules and proteins. Aptamers apply to therapeutic agents that can capture specific target molecules with stability, non-toxicity, and non-immunogenicity.
Hydroxyapatite is a typical material used for bone grafting due to its structural and chemical similarity to the hard tissue. Hydroxyapatite is also used to promote bone regeneration by modifying the surfaces.
In this study, we have developed the novel aptamer conjugated hydroxyapatite (Apt-HA) that promotes bone regeneration and angiogenesis. 3R02 bivalent aptamer, which specifically captures the vascular endothelial growth factor (VEGF) protein, was used. Certain cross linkers were used to graft the aptamers on the surface of hydroxyapatite, which was identified by the FT-IR spectrometer. Apt-HA was tested for its ability to capture VEGF protein, which was observed under a fluorescence microscope using FAM-labeled aptamers. The result showed that Apt-HA has higher capturing ability to VEGF protein rather than original hydroxyapatite.
In summary, Apt-HA can be used as advanced bone graft by promoting bone regeneration and angiogenesis.
