In this study, phospholipid-immobilized CdSe/ZnS core-shell quantum dots (PCZQDs) were prepared by a coupling reaction of trioctylphosphine oxide-coated CdSe/ZnS core-shell quantum dots (CZQDs) with DSPE-PEG (2000) and carboxylic acid-terminated PEG-2-PE (PCZQDs). Then, herceptin was introduced to the surface of PCZQDs (HPCZQDs) to enhance the antitumor effect of chemotherapeutic agents without increasing their toxicity. The surface properties of PCZQDs and HPCZQDs were characterized by fourier-transform infrared (FT-IR), electron spectroscopy for chemical analysis (ESCA), UV-vis spectrophotometer (UV-vis), transmission electron microphotographs (TEM), dynamic light scattering (DLS) and zeta potential. To evaluate the cell compatibility and cytotoxicity of PCZQDs and HPCZQDs, human breast cancer cells were cultured in the presence of Q-dots. Interaction of HPCZQDs with cells was also observed by confocal laser scanning microscopy (CLSM). |
