Enhanced cancer chemo-immunotherapy using nano-micelled generating dissolving microneedles containing paclitaxel and TLR7/8 agonist |
초록 |
Efficient drug delivery with microneedle (MN) of low dose chemical therapy agent immune-modulators adjuvants-resiquimod (R848) and paclitaxel (PTX) was synergized to demonstrate anti-cancer effect and activation of antigen-presenting cells (APCs), which thereafter induce anti-cancer memory effect. Paclitaxel treatment was speculated to cause cancer cell death which should lead to production of malignant tumor-specific antigens. R848 was used as an adjuvant and was anticipated to induce activation and maturation of immune cells for eliciting anti-cancer immune response. Here, Pluronic F127 based DMNs in situ generate nano-micelles (NMCs) upon their dissolution after subcutaneous injection. The capability of concurrent delivery of two agents-loaded NMCs was researched by watching the induced secretion of pro-inflammatory cytokines which are considered for anti-cancer activity and the expression of maturation markers (CD40, CD80) which elicit a much stronger immune stimulation effect. |
