Arbidol, which is applied to inhibit viral infection including influenza virus in Russia and China, has been revealed its inhibitory mechanism recently. Arbidol stabilizes the trimeric structure of hemagglutinin (HA) of non-resistant influenza virus by binding with the hydrophobic pocket which size is 870~1150 A, while the corresponding part in the resistant one is 90~150 A. Based on this fact we determined the combination of arbidol-fluorescent dye that would bind with the pocket in HA from the crystal structure of HA in complex with H3N2 and H7N9 viruses, finding that Arbidol conjugated with Rhodamine 123 (Arb-Rho123) can be applied for the detection. Then Arb-Rho123 is synthesized by EDC-NHS coupling reaction, and it is exposed to the influenza virus. Indeed, the intensity of Arb-Rho123 was decreased to 55% over 2.5x10-8 M of Arb-Rho123 with H3N2 compared to the control one. |
