제목 Smart magnetic nanospheres with tumor-specific ability for MR molecular targeting
초록 We have investigated the use of antibody–conjugated smart magnetic nanospheres as a potent targeted magnetic resonance (MR) contrast agent for a therapeutic or a diagnosis agent. Superparamagnetic iron oxide nanoparticles (IONs) were synthesized by a chemical coprecipitation method. IONs were first encapsulated with PLGA by using an emulsification-diffusion method. And then, Herceptin(the first therapeutic antibody approved for the treatment of HER2 positive metastatic breast cancer) was conjugated to the encapsulated nanoparticles by the chemical reaction between the succinimidly ester group of encapsulated nanoparticles and the amine group of mAb Herceptin. FACS and MR molecular imaging revealed that the overexpressing cell of HER2/neu showed a stronger contrast enhancement than the non-expressing cells of HER2/nue and signal intensity of in vitro MR imaging increased as the concentration of mAb Herceptin increases.
