An animal cell exerts traction forces to a substrate covered with extracellular matrix (ECM) to sense the rigidity of the substrate and migrate over the substrate. Traction forces that occur during migration are generated by a dynamic tugging of filamentous actin (F-actin) cytoskeleton. Filamin A is a protein cross-linking to actin molecules and thus considered involved in generating traction force. However, there are still no quantitative data showing the importance of Filamin A in traction force. Recently, traction forces have been measured by traction force microscopy technique using soft pillar made by soft material such as polydimethyl siloxane (PDMS). In this study, we measured traction forces of brain tumor cell line (U-87 MG) and its Filamin A-depleted cells using soft pillars coated with fibronectin. Average traction force per single pillar of wild type U-87MG cell is 4.15 nN, while Filamin A depleted cells has 3.63 nN. This is about a 15% decrease in traction force in the depleted cells, compared with the wild type cells. Our results suggest that the cross-linking of actin molecules Filamin A has modest effect on traction force generated by U-87 MG. |
