Chitosan has been appeared to be useful in biomedical applications due to its electrostatic interactions with anionic glycosaminoglycans (GAG). On the other hand, HA is a mucopolysaccharide found in various types of tissues and its immunoneutrality makes it an excellent building block for biomaterials to be employed for tissue engineering. Both of them are very attractive in hydrogel form, which has been used as scaffold for bone-tissue regeneration. However the hydrogel has lacking in osteoblast activities which can be modified by introducing Bioglass particle. BMP-2, like other bone morphogenetic proteins, plays important role in the development in bone regeneration. In this work, porous hydrogel scaffold of Ch-Hya-MP2-Bg was prepared by vigorous mixing, freezing and lypholization methods. Several experiments were undertaken to analyze the physical properties of the scaffold such as SEM, EDX, XRD, FT-IR, Swelling ratio, water uptake, compressive strength and porosity. MTT and confocal imaging as an in vitro experiment showed that loading of Bg particles increased the MC3T3-E1 preosteoblast cell viability which was increased further by introducing BMP-2. Thus the composite scaffold has a great future in bone tissue regeneration. |
