Label-free detection with ligand immobilization on SERS-encoded and polydiacetylene-coated beads |

Multiplex protein detection technology is a powerful tool to investigating the interaction of protein and protein. Since the efficiency and convenience, encoded bead was usually used for this detection method. However, high-throughput detection of encode bead is still challenging because of the difficulty of binding between labeled secondary protein and target protein. In this study, we introduced biotin to the polydiacetylenecoated surface-enhanced Raman scattering-encoded beads (PDA–SERS beads) to detect target protein model (streptavidin) in the PDA-SERS bead system. The fluorescence intensities of biotin-conjugated PDA–SERS beads were increased by the increasing concentration of streptavidin. We could detect streptavidin with detection limit of 2ⅹ10-8 M throughout the fluorescence intensity without additional labeling step. |
