제목 | Development and applications of cell-free protein synthesis technology |
초록 | Exponential increase in newly identified DNA sequences from various sequencing projects has resulted in the need for high-throughput protein expression that is hard to be achieved by cell-based expression methods. When compared to cell culture methods that involve gene cloning and cell culture steps, a cell-free synthesis system has substantial advantages in terms of speed and flexibility for protein production; therefore, it is more suitable for high-throughput protein production. The productivity and economics of cell-free protein synthesis have also been improved remarkably during the past decade, and production of milligram amounts of protein is now being reported. This presentation will cover the biochemical and genetic approaches we have undertaken to improve the productivity and economic efficiency of cell-free protein synthesis systems. Applications of the cell-free synthesis system in the areas of protein engineering, enzyme screening, and diagnostics will also be discussed. |
