제목 Application of polymeric micelles containing Fe2O3 NPs for protein purification
초록 We report a new system for protein purification based on magnetic properties of Fe2O3 NPs and selective binding of Ni-NTA complex with histidine tagged protein. We first synthesized the amphiphilic polymers, nitrilotriacetic acid (NTA)-end functionalized polystyrene by ATRP using N',N'-Bis[(tert-butyloxycarbonyl)methyl]-N''-Bromoisobutyryl-L-lysine tert-Butyl Ester as an initiator. When the amphiphilic polymer and superparamagnetic iron oxide nanoparticles were dissolved together in THF, the addition of water caused the spontaneous formation of polymeric micelles, resulting in the successful encapsulation and aqueous dispersion of NPs. After complexation of Ni2+ with NTA on the surface of the polymeric micelle containing NPs, the specific binding between Ni-NTA complex and His-tagged green fluorescent protein was studied. We confirmed the selective separation of His-tagged protein using the micelles containing Fe2O3 NP by SDS-page and Fluorescent emission.
