Dual growth factor delivery using core PLGA-shell alginate microcapsule for angiogenesis |

Growth factors (GFs) are very critical in tissue regeneration using stem cells. Presently a precise release control of multiple GFs is still challenging. This study investigates core-shell microcapsules (MCs) that deliver two angiogenic factors: VEGF and PDGF. MCs were manufactured using an electrodropping system and PLGA emulsion and alginate were employed as core and shell material, respectively. These MCs are supposed to sequentially deliver each GF with a concentration gradient. Release profile of GFs from MCs was tunable using either different MW of PLGA and/or layer-by-layer assembly. Furthermore MCs proved effective in stimulating the angiogenic responses of endothelial cell in vitro, as demonstrated by angiogenic sprout and capillary-like structure. The proposed dual GFs delivery MCs can help us better understand how cells respond to a concentration gradient of multiple signals that is available over a long-term period of time. |
