Gene silencing using siRNA can be a new potent strategy to treat various diseases and also adjusted to the treatment of skin disorders. For adjusting to the skin, however, it is limited due to its rapid degradation and poor cellular uptake. Thus, we designed specific lipid nanoparticles for the delivery of siRNA into the skin and evaluated the potential of the particles. we used 1,2-dioleoyl-3-trimethylammonium-propane (DOTAP) to prepare a nanocarrier for delivering siRNA against GAPDH, a housekeeping gene, and compared its efficiency with lipofectamine as another lipid carrier through some bioanalysis. In addition, a dermal transport assay is designed and implemented to evaluate the penetration and delivery efficacy of siRNA in pig skin using lipid nanocarriers. We plan to develop these nanoparticles to improve the permeability of skin structures for the enhanced delivery of siRNA-loaded nanocarriers in the near future. |
