제목 | Engineering versatile photochemically triggered nanoparticles for targeted tumor therapy |
초록 | Nanoparticle (NP)-based tumor therapy have attracted much attention due to their tunable physical/chemical characterizations and their outstanding drug loading efficiency. Despite their strengths of the systems, there are some fundamental limitations that reduced intracellular delivery and endo/lysosome escaping. To design an advanced NP-based tumor therapy, we prepared pH sensitive polymeric NPs combined with photodynamic therapy (PSNP). A versatile NP is formed by self-assembly of a photosensitizer, pH-sensitive moiety conjugated to polysaccharide as tumor targeting agent and doxorubicin. The pH responsiveness of PSNP leads to disassembly of PSNP owing to the protonation of pH-sensitive moiety, which causes DOX release. Upon light irradiation, entrapped photosensitizers controlled oxygen molecules to produce singlet oxygen (1O2). Confocal microscopy and competitive cellular uptake studies revealed receptor-mediated endocytosis and endosomal escape capability. |
