Optimization of scaffold and hydrogel-seeding method for vascular tissue engineeering |
초록 |
This study was focused on a hydrogel-seeding method to improve cell adhesion and proliferation in tubular porous scaffolds for vascular grafts application. Tubular scaffolds were fabricated by an extrusion-particulate leaching method using a biodegradable elastic polymer, poly(L-lactide-co--caprolactone) (PLCL) (50:50, Mn 1.58105). we polished the outer lumen of scaffolds with sandpapers to remove the closed pore for the improved infiltration of cells-hydrogel mixture into the inside of scaffold. In addition, we used a vacuum seeding method to infiltrate cell-hydrogel mixture into scaffolds. Vascular smooth muscle cells (VSMCs) were dispersed in collagen hydrogel and then seeded into the tubular PLCL scaffolds having various pore sizes 50-100μm, 100-200μm, and 300-500μm, respectively. Especially, the cell proliferation efficiency improved by using the hydrogel-seeding method as compared with by using an established method. |
