제목 In Vivo Evaluation of Simvastatin by Self-Emulsifying
초록 A self-emulsifying drug delivery systems (SEDDS) was developed to improve the oral bioavailability of poorly water soluble drugs because of its ability to improve drug solubilization and potential for enhancing absorption in the gastrointestinal tract, caused by surfactant-induced permeability. Simvastatin, a poorly water soluble drug and widely used for the treatment of hypercholesterolimia, was incorporated into the SEDDS. Optimized formulations for bioavailability assesment were selected by evaluating the solubility and microemulsion existence range after emulsifying with varying compositions of oil and surfactant-cosurfactant mixtures, and by determination of particle size of the resulting emulsion. The absorption profile from SEDDS was compared to simvastatin tablet (Zorco?) by the administeration as prefilled hard gelatin capsules to fasted beagle dogs. The SEDDS formulations of simvastatin showed improvement in absolute oral bioavailability relative to previous data of commercial simvastatin tablet formulation. These data indicate the utility of dispersed self-emulsifying formulations for the oral delivery of simvastatin and potentially other poorly absorbed drugs.
