Polymeric Hydrophobic Gold Nanoparticles for X-Ray Computed Tomography Imaging Contrast Agent |

Gold nanoparticles (GNPs) can absorb and scatter the electromagnetic radiation induced from x-ray and enhance their intensities because of strong electric fields at surface. In this study, we synthesized polymeric hydrophobic gold nanoparticles (PHGNs) to serve as nano-probes for x-ray computed tomography imaging contrast agent. First, hydrophobic GNPs were synthesized by reduction of HAuCl4 in the presence of sodium borohydride and dodecanethiol. Water dispersion PHGNs were then finally obtained by the encapsulation of hydrophobic GNPs with sodium dodecyl sulfate (SDS) as an amphiphilic surfactant by nano-emulsion method. The morphology of PHGNs was evaluated by transmission electron microscopes and revealed that hydrophobic GNPs were successfully encapsulated by SDS. The chemical structure of the synthesized PHGNs was confirmed by FT-IR. The surface charge and size distribution of prepared PHGNs were analyzed using dynamic light scattering. We performed a MTT assay to examine the cytotoxicity of PHGNs using NIH3T6.7 cell (human breast cancer cell). We calibrated the x-ray absorption of the PHGNs to investigate CT contrast efficacy of PHGNs. Consequently, these results demonstrate that synthesized PHGNs have highly versatile biomedical characteristic such as optical and x-ray computed tomography imaging contrast agent. |
