We synthesized block copolymers of poly(ethylene glycol) and poly(L-serine) bearing poly(D,L-lactide) (PLA) side chains. In the synthesis, poly(ethylene glycol)-b-poly(L-serine) was first prepared by ring opening polymerization of N-carboxyanhydride of O-(tert-butyl)-L-serine using methoxy-poly(ethylene glycol)-amine as an initiator. After removal of tert-butyl groups with hydrogen bromide, PLA was grafted to poly(L-serine) via ring opening polymerization of D,L-lactide in the presence of tin(II) 2-ethylhexanoate as a catalyst. The drug loaded nanoparticle of the copolymer was prepared by an emulsion-solvent evaporation method using 1-aminopyrene as a model drug. The nanoparticle was characterized in terms of size, surface charge, and drug release profile. |
