제목 | Evaluation of antitumor activity for self-microemulsified BCNU-loaded PLGA wafer against 9L gliosarcoma cells in in vitro |
초록 | Interstitial therapy using biodegradable polymeric device loaded with anticancer agent can deliver the drug to the tumor site in high concentration and hence increase therapeutic efficacy. 1,3-bis(2-chloroethyl)-1-nitrosourea(BCNU) is the most commonly used chemotherapeutic agent for brain tumors. But it is unstable to heat and light, and also has a half life of 20min in vitro and less than 15min in vivo by hydrolysis. Self-microemulsion system was developed for enhancement of BCNU half-life from 45min to 130min in vitro. The self-microemulsified BCNU was incorporated into PLGA wafer as a new polymeric implant. Harvested cells were plated in 6-well flat-bottomed plates (2X104 cells/ml). After 24h, free BCNU-loaded PLGA disk or self-microemulsified BCNU-loaded PLGA wafer was placed in contact with the attached cells. After incubation for specific time period, the cytotoxic activity was evaluated by using sulforhodamine B assay (Abs, 520 nm). The results of the experiments were expressed as treatment over control (T/C) values. Cytotoxicity of self-microemulsified BCNU/PLGA wafer against 9L gliosarcoma cells was higher than free BCNU wafer due to the longer half-life of BCNU.
