Thermosensitive hydrogels, high water-swellable polymer materials, are useful in a wide variety of biomedical applications, including responsive injectable materials and tissue engineering matrices. Poly(ε-caprolactone) (PCL) as a well-known biodegradable polymer in biomedical field has good biocompatibility and sufficient physical properties. Heparin, a representative anticoagulant can be modulated the bioactivity of heparin binding growth factor (bFGF, VEGF and etc.) and cell-related cytokines through different mechanism. The objective of the present study is to develope multivalent PCL-heparin conjugate using tetrafunctional PEO-PPO block copolymer (Tetronic®) for pharmaceutical and tissue engineering applications. Tetronic-PCL copolymer was synthesized by ring opening polymerization of ε-caprolactone with Tetronic® using stannous octoate. Then, heparin was coupled to Tetronic-PCL copolymer by EDC / NHS chemistry. Tetronic-PCL-heparin conjugate was characterized by FT-IR, NMR, GPC, the sol-gel transition behaviors and Dynamic Light scattering (DLS). The characterization of immobilized heparin was estimated with toluidine blue assay and activated partial thromboplastin time (APTT) test. Obtained results suggest that heparinized injectable polymers have great potentials for various biomedical applications. |
