Prevention of inflammation induced by atRA-loaded microspheres with concurrent administration of dexamethasone |

In this study, microspheres containing all-trans retinoic acid were subcutaneously injected into rats and dexamethasone was orally administered to prevent inflammation induced by injected atRA-loaded microspheres. Therefore, we hope that the metabolism of atRA can be suppressed due to the prevention of inflammation induced by atRA-loaded microspheres when dexamethasone is concurrently administered. Many fibroblasts around microspheres were observed at implant sites of atRA-loaded microspheres. But, dexamethasone effectively inhibited the fibroblast proliferation compared to atRA-loaded microspheres treated group alone. The atRA plasma concentration in atRA MS treated group was lower than those in co-treated group of atRA MS and dexamethasone. Therefore, we concluded that co-administration of dexamethasone could effectively prevent fibroblast proliferation and severe collagen deposition acted as obstacles of atRA systemic delivery resulting in the increase of atRA plasma concentration. |
