제목 Application of paclitaxel-loaded PLGA wafers using self-microemulsion system for local therapy
초록 The aim of this study is to develop for suitable formulation to make optimal paclitaxel-loaded PLGA wafers for implantable matrix form using self-microemulsifying system. Self-microemulsifying drug delivery system (SMEDDS) is isotropic mixtures of an oil, surfactant, cosurfactant and drug. Paclitaxel is one of the best antineoplastic drugs with demonstrated cytotoxic activity against a number of tumors including breast and ovarian cancer and formulated in a mixture of 50:50% (v/v) Cremophor EL® and dehydrated ethanol for injection due to extremely low aqueous solubility. However, Ctemophor EL® has well known side effects such as hypersensitivity reactions. Therefore PLGA and self-microemulsified paclitaxel without Cremophor EL® were mixed, sieved and fabricated by compression molding method for implantable matrix form. The in vitro release study was performed in 1% SDS solution at the constant temperature (37℃) under sink condition. From the result of in vitro release test, we could induce the desirable drug release patterns and enhancement of bioavailability by means of the ratio of oil and surfactant. In conclusion, the use of biodegradable implant matrix of self-microemulsified paclitaxel without Cremophor EL® maybe providing the bioavailability of paclitaxel and decrease of side effects.
