Efficient Intracellular Delivery in Cancer Cells by Reactive Oxygen Species (ROS)-Responsive Polymer |

The high intracellular oxidative stress in a cancer cell is a biologically relevant stimulus to be utilized for efficient intracellular delivery. We designed and synthesized reactive oxygen species (ROS)-responsive, cationic polymer to achieve efficient intracellular gene delivery in prostate cancer cells. DNA complexed with PATK was efficiently released via exposure to high levels of ROS in prostate cancer cells. As a result, higher gene transfection efficiency of DNA/PATK polyplexes compared to their non-degradable counterparts was achieved. Further conjugation of cancer-targeting peptide to the the polymer significantly increased its cellular uptake in prostate cancer cells, leading to cancer-targeted gene delivery. This study demonstrates that ROS-responsive polymer combined with a cancer-targeting peptide is an efficient and cancer-targeted carrier. |
