Effective chondrogenic differentiation of stem cell in RGD-grafted Pluronic F127 hydrogel |

Thermogelling Pluronic F127 hydrogel approved by the FDA has been studied extensively for drug delivery system and tissue engineering as a biomaterial for human applications. For improving cell adhesion and biodegradability FGM-RGD was synthesized by covalent bonding of RGD peptides to F127-glycolide-4-methacryloxyethyl trimellitic anhydride (FGM), where Pluronic F127 derivatives have novel temperature-sensitive properties. These polymers were characterized by 1H-NMR, FTIR, GPC, and elemental analyzer. In the cell viability study, the optical density (OD) of cell in FGM gel was resulted in very low value. It was revealed from cell counter kit-8 (CCK-8) that the toxicity increased as the content of APS/TEMED increased, and resulted in inhibiting the cell viability of chondrocytes. In order to enhance cell viability in hydrogel, COOH group-blocking FGM and RGD-grafted FGM will be synthesized and characterized. |
