Injectable, porous, biodegradable microspheres for stem cell tissue engineering |

We studied the use of biodegradable porous microspheres as cell cultivation carriers and injectable scaffolds for tissue regeneration. Highly open-porous microspheres were fabricated using poly(D,L-lactide-co-glycolide) by a novel single emulsion-phase separation technique. The porous microspheres were used as scaffolds to form cellular aggregates of human mesenchymal stem cells. The cellular aggregates were cultured for 3-D differentiation into adipocytes. To assess the potential of cellular aggregates as injectable adipose tissues for tissue regeneration, aggregates were implanted in vivo by injection. Only the differentiated cellular aggregates resulted in functional tissues expressing the adipocyte-specific phenotype. |
