Fabrication of porous poly(lactic-co-glycolic acid) microspheres |

Double emulsion solvent evaporation method has been popularly used to encapsulate protein drugs within biodegradable poly(lactic-co-glycolic acid) (PLGA) microspheres. We fabricated highly porous biodegradable polymeric microspheres by solvent evaporation technique using a polymer blend composed of water insoluble polymer (PLGA) and water soluble polymer (Pluronic F127) as a porogen. The pores are uniformly distributed and well inter-connected on the surface and within inside of resultant PLGA microspheres. These pores could be closed by the treatment of various water-miscible solvents. Gradual morphological change was observed from a pore-opened state to a pore-closed state. These open pore PLGA microspheres could be used likely for sustained delivery of bioactive macromolecules |
