Growth Factor-Loaded Porous PCL Beads as an Injectable BioactiveBulking Agent for the Treatment of Urinary Incontinence |
초록 |
Urinary incontinence, which is defined as the involuntary loss of urine, may be caused by uretheal hypermobility and/or intrinsic sphincter deficiency. Injectable bulking agents have been used in the treatment of urinary incontinence. However, injection volume decrement caused by particle migration and resorption in the body has been a main problem. In this study, we prepared growth factor-loaded porous polycaprolactone (PCL)/Pluronic F127 beads as an injectable bulking agent. The Pluronic F127 chains exposed onto the PCL bead pore surfaces were used to bind heparin and the following growth factors (bFGF or VEGF) which may improve the sphincter muscle function around the urethra by the induction of smooth muscle cells. The morphologies and growth factor release behaviors of the beads were investigated by SEM and ELISA kit, respectively. The induction potential of smooth muscle by the growth factor-loaded porous beads was also evaluated using muscle-derived stem cell as a model cell. |
