제목 Release Profiles of Poorly Water-soluble Drug from Biodegradable Microspheres for The Treatment of Osteoporosis
초록 The poly(lactide-co-glycolide) (PLGA) due to its controllable biodegradability and excellent biocompatibility is one of the most significant candidates for the study of biodegradable polymeric controlled release. Ipriflavone (IP) stimulates proliferation and differentiation of osteoblast and also enhances calcitonin secretion in the presence of estrogen. IP (poorly water-soluble drug) loaded PLGA microsphere (MS) was prepared by using conventional O/W solvent evaporation method. The different morphology and size distributions can be changed by various factors in the preparation procedure such as molecular weight (8K, 20K, 33K and 90K g/mole) polymer concentration (2.5, 5, 10 and 20%), surfactant types (SDS, PVA, gelatin and Tween 80), initial drug loading (5, 10, 20 and 30%), stirring speed (250, 500 and 1000 rpm) and evaporation temperature (25, 35 and 45℃). In vitro drug release behaviors of IP were determined by high perfomance liqud chromatography (HPLC). These results suggested that IP-loaded PLGA MS may be used as carrier in controlled delivery system for the treatment of osteoporosis.
