제목 Study on Geometric Effects of BCNU-loaded PLGA Wafer on BCNU Release
초록 Interstitial therapy using biodegradable polymeric device loaded with anticancer agent can deliver the drug to the tumor site in high concentration and hence increase therapeutic efficacy. 1,3-bis(2-chloroethyl)-1-nitrosourea(BCNU) is the most commonly used chemotherapeutic agent for brain tumors. The design of implantable device is regarded as an important factor for the efficient delivery of antitumor agent to a tumor. The purpose of study is to investigate the release pattern of BCNU from various dimension changes of the wafers, such as weight, diameter, thickness, volume, and surface area of wafers. The BCNU-loaded PLGA wafers containing 3.85%, 10%, 20% and 30% of BCNU were prepared in various shape (diameter of 3mm, 5mm and 10mm, thickness of 0.5mm, 1mm and 2mm). In vitro drug release profile of BCNU-loaded PLGA wafers were analyzed by high performance liquid chromatography (HPLC) assay. Moreover, the effects of various geometric factors were confirmed by investigating mass loss, morphology, and molecular weight change of the wafer as well as pH change of release test media.
