제목 Bone Formation of Bone Marrow Stromal Osteoprogenitor Cells in Ipriflavone Loaded PLGA Scaffold: Characterization and in vivo Study
초록 Ipriflavone (IP), a non-hormonal isoflavone derivative, has been shown to interfere with bone remodeling by inhibiting bone resorption and stimulating bone formation. IP-loaded PLGA scaffolds were prepared by novel solvent casting/particle leaching and emulsion freezing drying methods. Characterizations of IP-loaded PLGA scaffolds were analyzed by scanning electron microscope (SEM), mercury porosimetry, X-ray diffractometry (XRD), differential scanning calorimetry (DSC) and so on. Released amount of IP from PLGA scaffold was assayed by high performance liquid chromatograghy (HPLC).
IP consistently increased the amount of 45Ca incorporated into the cell layer by bone marrow stromal osteoprogenitor cells (BMSOCs), and stimulated mineralization of BMSOCs assessed by von Kossa staining. We studied the role of BMSOCs reconstituted IP-loaded PLGA scaffolds in athymic nude mice. Differential cell morphology was observed by H&E staining and SEM.
