제목 Implantable Pamidronate-loaded PLGA Disk for Treatment of Bone Resorption
초록 Bone resorption is mostly reponsible for serious complication caused by aural cholesteatoma. Recent researches have been aimed at non-surgical therapy for the localized bone resorption in cholesteatoma. Pamidronate disoum (3-amino-1-hydroxypropane-1,1-bisphosphonate, APD) is one of bisphosphonates which have been used to inhibit bone resorption . In this study, APD is chosen as a model drug for its lower autotoxicity than other bisphosphonates and incorporated into PLGA for homogeneous mixing and fabricated into disk. The effect of manufacturing parameters on release of APD from disk, such as initial drug loading, thickness, diameter, and water-soluble excipients, were investigated. In vitro release profile of APD from disk was obtained by HPLC analysis after derivatization of APD with 1-naphthylisothiocyanate. In vivo release of APD implanted in mongolian gerbil was investigated to compare the in vitro release of APD. Crystallinity of APD, molecular weight change, and morphology of APD/PLGA disk during release test period were also investigated.
