In our previous study, poly(4-vinyl imidazole)(P4V) exhibited higher transfection efficiency with lower cytotoxicity than polyethylenimine(PEI) owing to pH-responsive endolysosome escape. But the drawback of P4V is insoluble in aqueous solution above at pH 6. In this study, we modified P4V with gluconic acid (P4V-GA) to impart solubility at physiologicical pH. The P4V-GA was synthesized by amidation of the -amine of P4V with urocanic acid by using an 1-ethyl-3-(3-dimethylaminopropyl) (EDAC)/ N- hydroxysuccinimide (NHS) condensation system. The P4V-GA/DNA complex formation was confirmed by electrophoresis on 1.0 % agarose gel. Particle sizes were determined using a light scattering spectrophotometer (ELS 8000, Otsuka Electronics, Ltd., Osaka, Japan). Cytotoxicity test was used by Cell Proliferation Assay Kit ( Promega Co.). The in vitro transfection experiment was checked by luciferase assay with several cells. The P4V-GA will be expected to be useful for clinical application of gene therapy with improving the transfection efficiency and cell viability.
