Polysaccharide-based hydrogel has been widely used as a vehicle for oral insulin delivery due to its mucoadhesiveness and biocompatibility. However, its applications have been hindered by low delivery efficacy in the systematic circulation and non-controlled release. Additionally, to endow glucose-responsive characteristic to drug delivery systems, PBA moiety was commonly used. In such system, with the increase of glucose level, PBA moiety can be converted from hydrophilic to hydrophobic by a negative ionization mechanism Herein, we suggested that Glycol chitosan(GC)/Sodium alginate(SA)-poly(L-glutamic acid-co-N-3-L-glutamyl amido phenylboronic acid)(PGGA) double-layered nanogel exhibits glucose-responsive characteristic and biocompatiblity. Therefore, the double-layered nanogel may be a potential platform for oral insulin delivery system. |
