Monoolein (MO) cubic phase, mainly used as drug carriers, was developed by immoblizing hydrophobically modified glucose oxidase (HmGOD) in the lipid matrix and containing acidic proteinoid in the water channel to prepare pH-dependent and glucose-sensitive system. Firstly, Glutamic acid and Isoleucine (98.5:1.5 in molar ratio) was used to prepare acidic proteinoid which would be condensed when pH < pI. As a result, the release of insulin loaded in the cubic phase would be promoted in acidic condition. Moreover, when the concentration of glucose is much higher, it reduced pH in water channel of cubic phase due to HmGOD interaction with glucose, which further affected acidic proteinoid, so that the release of insulin from cubic phase was higher. Therefore the developed cubic phase was potential for both pH-dependent and glucose-sensitive drug delivery system. |
