Mode of action of synthetic antimicrobial peptides with cell selectivity |

Emergence of multidrug-resistant bacteria has been raised a limited antibiotic-treatment and demanded a new antibiotic. Antimicrobial peptide(AMP) has different action mechanisms, which are physical perturbation and disruption in microbial membrane, compared with antibiotics. We rationally designed synthetic AMPs by using lysine, isoleucine, typtophan, arginine and glutamic acid. The synthetic peptides showed a potent antibacterial activity for planktonic cells in Pseudomonas aeruginosa and Staphylococcus aureus, drug-resistant bacteria isolated from patients. They are non-toxic against human erythrocytes and mammalian cells. The results are able to provide basic information in designation of synthetic peptides as candidate of AMPs.
