Glutamate decarboxylase (GAD) is an enyzme that catalyzes the decarboxylation reaction of glutamate by which gamma-aminobutyric acid (GABA) is synthesized. The reaction procedes with the concomitant consumption of a proton, which increases pH of the medium. Since the enzyme showed cooperative structural changes into nonreactive ones upon pH increase, efforts were endeavored to keep the active structure over a broader range of pH, especially toward alkalinity. Here I present the effects of site-directed mutations on the enzyme at positions 89, 465, and the ultimate residue in a combinatorial manner. |
