Collagen/cell incorporating elastic scaffold using pulsatile perfusion bioreactors in the development of a small-diameter vascular graft |
초록 |
This study reports on the effects of dynamic culture system and Collagen/ smooth muscle cells (SMCs) mixtures on the cellularity and composition of smooth muscle tissues engineered using biodegradable elastic scaffolds of 4 mm inner diameter and 5 cm length. Rabbit aortic SMCs were infiltrated with collagen solution in tubular PLCL scaffolds under vacuum to form collagenous gel and subjected to pulsatile strain and shear stress by culturing them in pulsatile perfusion bioreactors. The collagen/SMCs incorporating elastic scaffold under pulsatile flow conditions promotes matrix deposition leading to the development of tissue-engineered vascular constructs and induce SMC to have more uniform cell distribution. A collagen/SMCs incorporated scaffolds under pulsatile perfusion conditions can be used as a scaffold for tissue engineering to facilitate small-diameter vascular-tissue formation. |
