Hyaluronic acid(HA) has been applied widely in the medical field as a drug delivery system, tissue engineering scaffold, and anti-adhesion agent. Photoreactive azidophenyl hyaluronic acid derivative was synthesized to immobilize proteins such as epidermal growth factor and bone morphogenetic protein. The photosensitizing group, 4-azidoaniline, was introduced to the carboxyl group of hyaluronic acid by a water soluble carbodiimide, EDC hydrochloride. To characterize the synthesized Az-HA, FT-IR, 1H-NMR and UV-visible absorbance spectroscopy were performed. We demonstrated protein immobilization by using fluorescein isothiocyanate conjugated albumin on Az-HA derivative surfaces that were patterned with micro scale. Az-HA derivatives were performed to cytotoxicity and cell proliferation tests to confirm biocompatibility. Because it is nontoxic to the cells, we suggest that the azidophenyl hyaluronic acid derivative could be applied to the medical field as a base material. |
