Biomarkers such as DNA, RNA, protein represent various biological conditions. Therefore, a simple, rapid and sensitive biomarker sensing method is currently needed for early and reliable diagnosis and development of point-of-care testing. Here, we report a target-assisted proximity proteolysis reaction that can recognize specific biomarkers and generate signal as a result. In this reaction, protease and zymogen were designed to recognize different positions in a target, thereby the target will enhance a proteolysis and generates a detectable signal. The developed nucleic acids sensing method can detect pico-molar range of nucleic acids and discriminate single-base mismatch within one hours. Sensing method for protein biomarkers also showed similar results of sensitivity and selectivity. The results demonstrated the feasibility of the proximity proteolysis reaction as a diagnostic platform for detecting various types of biomarkers. |
