Lipases (EC 3.1.1.3) that catalyze the hydrolysis of triglycerides in vivo are of great value for the modern chemical and pharmaceutical industries. Lipase can be used as an efficient catalyst for biodiesel(FAME ; Fatty Acid Methyl Ester) production. In this study, we used immobilized lipase for the production of biodiesel from oil. Cross-linking method was used for immobilization because it serves stronger binding and stability than other methods. The objects of this study are to develop a suitable immobilization method of lipase and its application of biodiesel production from oil with suitable activity and stability. Hydrogen peroxide concentration of 35%(v/v) was determined for an the most effective pretreatment. To activate carrier, 10% 3-APTES was used. As a coupling reagent, 2% glutaraldehyde wass treated. About 11.2U/ml of lipase was loaded to carrier. 92% of protein was attatched and 28.9% of lipase activity was recovered. This value was comparable to the results by other studies. After 20 times reusing, it maintained about 80% activity. It was prominent result compare to other studies. |
